Chromium Xo: Everything You Need to Know About 10x Genomics' New Low-Cost Single-Cell Solution
Discover how the Chromium Xo offers a cost-effective entry into single-cell research while providing flexibility for future upgrades.
When you hear about 10x Genomics' new low-cost instrument, the Chromium Xo, you might think it’s a completely new device. It’s not. The Chromium Xo is part of the Chromium X series, just like the iX and X models. The machine is the same; the difference is in the software—and the price.
Here’s a breakdown of what you need to know:
The Chromium Platform
The Chromium platform is 10x Genomics' machine for single-cell sequencing. You might know about the older Chromium Controller or the newer Chromium X series. The Controller will be discontinued soon, but 10x Genomics introduced the Chromium X in 20211 as its replacement, offering multiple versions (iX, X, and now Xo). See Fig.1
What is the Chromium Xo?
The Chromium Xo is a budget-friendly version of the new Chromium, designed to run the GEM-X Single Cell Gene Expression v4 assays. It costs about $25,000 (US price)2 because it limits certain features. However, it has no limit on the number of runs, making it a great starting point for labs focusing on 3' RNA assays.
What Can You Do?
Use cell suspensions, fresh or frozen tissues
Use cells or nuclei
Run up to 8 samples per experiment
Study RNA expression (3’ region) using GEM-X Single Cell Gene Expression v4
Analyze protein expression using a 3’ barcode kit and Biolegend TotalSeq-B antibodies
What Can't You Do?
Use FFPE samples (you’ll need the iX or X for that).
Run assays like immune profiling (5’), ATAC-seq, or multiome (you'll need the iX or X).
Process more than 8 samples per run (for up to 16 samples, you’ll need the X).
Upgrading the Chromium Xo
You can upgrade the Chromium Xo software to unlock all features, turning it into a full Chromium X but not an iX.
Why Consider the Chromium Xo?
If you’re holding off on buying a new Chromium machine due to budget concerns, the Chromium Xo offers a more affordable entry point into 10x Genomics single-cell technology. It’s perfect for labs that want to start with 3’ RNA assays and scale up later. If you only plan to do a few experiments, consider doing a collaboration, using a service provider or a free instrument assay.
For start-ups and core facilities, the Chromium Xo is a good investment if you plan to offer single-cell services. 10x Genomics is the gold standard in single-cell technology, so having it in your lab is essential for delivering high-quality research.
In any case, you should do the math—consider not just the instrument cost but also the ongoing expenses like reagents and maintenance.
The Bottom Line
The Chromium Xo isn’t a new instrument—it’s a cost-effective version of the Chromium X. It’s a smart way to access 10x Genomics technology. When you’re ready for more advanced assays, you can easily upgrade the software to unlock additional features. Check figure 2 for extra information about it.
