Tools for Single-Cell Sequencing Analysis in Bacteria and Yeast
B.SIGHT & GEXSCOPE® Microbial Single Cell RNA Library Kit
The most commonly used samples for single-cell sequencing experiments are human, 🐭, zebrafish, or 🪴 samples. However, interest in cells like bacteria or yeast is increasing. Over the last 2 months, I have already helped 3 👩🏻🔬 👨🏽🔬 design their projects. Today I will share some commercial tools, ideal if you want to do single-cell sequencing from bacteria and yeast.
B.SIGHT: Precision Cell Sorting for Bacteria or Yeast
B.SIGHT, from Cytena, is an advanced tool for isolating microbes one by one. Using high-resolution optical microscopy and specialized dispensing technology, it efficiently separates tiny cells like bacteria and yeast.
It works like this: a dispenser chip creates micro-droplets from the sample, while super-clear optics and real-time image analysis ensure only 1 cell goes into each well of a 96- or 384-well plate.
Pairing B.SIGHT with Cytena's I.DOT machine lets us perform the SMART-seq protocol using regents in tiny volumes like nanoliters. The SMART-seq protocol can also be done manually after the use of B.SIGHT. This method allows us to study full-length RNA transcripts from each individual cell. ‼️ Note: The original lysis buffer needs to be adapted for bacteria or yeast. We need stronger lysis when compared with eukaryotic cells.
GEXSCOPE® Microbial Single Cell RNA Library Kit: Unveiling Yeast Transcriptomics
Singleron's GEXSCOPE® Microbial Single Cell RNA Library Kit simplifies the process of creating RNA libraries from yeast cells. It needs to be used with Singleron’s microwell-based SCOPE® (Single Cell Omics Preparation Entity)technology, specifically the HD chips. These chips can be loaded either manually or with Singleron’s automated system, the Matrix Neo. Each chip can capture up to 30,000 individual yeast cells.
